In vitro model to study demyelinating disease using human OPC- and oligodendrocyte-like cells generated by deterministic cell programming
We have developed hiPSC-derived oligodendroglial cell models using opti-ox technology.
- ioOligodendrocyte-like cells resemble a pre-myelinating oligodendrocyte state, rapidly maturing to express MBP by day 8 in monoculture.
- MBP expression is further enhanced by optimising culture conditions, including seeding density and cAMP concentration, and by co-culturing with ioGlutamatergic Neurons.
- ioOPC-like cells can maintain their proliferative ability during maintenance phases, whilst retaining the capacity to mature into MBP-positive cells, by switching into maturation media.
Together, ioOPC-like cells and ioOLCs provide scientists easy access to physiologically relevant human oligodendroglial cell models, ideal for studying demyelinating and neurodegenerative diseases.
