This user manual is applicable to ioGlutamatergic Neurons  (wild type) and ioGlutamatergic Neurons disease model cells carrying mutations in HTT, TDP-43 (TARDBP), MAPT, PRKN, SNCA, APP, PSEN1, PINK1 and GBA.

The ioGlutamatergic Neurons mature rapidly into functional glutamatergic neurons after revival in the recommended, fully disclosed, medium. They are delivered in a convenient cryopreserved format and provide a homogeneous and reproducible model for human excitatory neurons.

The user manual provides full details for phases 1 and 2 in plate formats from 6- to 384-wells and includes a protocol for co-culture with astrocytes.

Induction

Carried out at bit.bio before distribution.

Phase 1 - Stabilisation (day 0 - 4)

ioGlutamatergic Neurons are revived at the user’s laboratory using the recommended medium.

Phase 2 - Maintenance (day 4 onwards)

Depending on assay requirements, the ioGlutamatergic Neurons can be cultured for different lengths of time in the maintenance medium.
