An mRNA-based approach enables efficient transfection of human iPSC-derived neurons and microglia, achieving ~60-95% efficiency with minimal toxicity and long-term sustained expression. High efficiency with high viability transfection requires systematic optimisation of seeding density, timepoint of transfection and mRNA/reagent amount.
