CRISPR-Ready ioGlutamatergic Neurons rapidly mature into functional glutamatergic neurons after revival in the recommended medium. These cells constitutively express Cas9 nuclease so users can perform single gene knockouts, pooled CRISPR screens or arrayed CRISPR screens. CRISPR-Ready ioGlutamatergic Neurons are delivered in a convenient cryopreserved format and provide a homogeneous and reproducible model of human excitatory neurons. 

The user manual provides full details for phases 1 and 2 in plate formats from 6- to 384-wells and includes protocols for gRNA delivery by either lentivirus transduction or lipid-based transfection.

Phase 1 - Stabilisation (day 0 - 4)
The CRISPR-Ready ioGlutamatergic Neurons are revived at the user’s laboratory using the recommended medium supplemented with doxycycline (96h) & DAPT (day 2-4) for sustained induction. Depending on assay requirements, the guide RNA can be delivered with lentiviral transduction or lipid-based transfection from day 1 to day 3 post thaw. 

Phase 2 - Maintenance (day 4 onwards)
Depending on assay requirements, the CRISPR-Ready ioGlutamatergic Neurons can be cultured for different lengths of time in the maintenance medium.
