This step-by-step protocol is recommended for assessing phagocytosis of pHrodo labelled bioparticles and amyloid-β 1-42 in ioMicroglia.

Microglia engulf and digest cellular debris, pathogens, and even synapses. This process, known as phagocytosis, is essential for maintaining brain health, removing harmful material, and supporting neuroplasticity. In Alzheimer’s disease, microglia are crucial in phagocytosing Amyloid-β and the fragment Amyloid-β 1-42 (Aβ42), which is a key component of amyloid plaques. This protocol describes how to assess the phagocytic capacity of ioMicroglia using three types of pHrodo-labelled bioparticles: E. coli, S. Aureus and Zymosan as well as a fluorescent HiLexa™ Fluor 488-labelled Aβ42 peptide.
