Co-culturing ioSkeletal Myocytes and ioMotor Neurons

This step-by-step protocol provides details on how to culture of ioMotor Neurons and ioSkeletal Myocytes in 24-well plates. It is also applicable to their respective ioDisease Model Cells.

Using this protocol, co-cultures have been successfully maintained for up to 30 days. Each cell type can be identified in co-culture by immunocytochemistry (ICC) using MAP2 (for ioMotor Neurons) and Desmin (for ioSkeletal Myocytes). Staining for the acetylcholine receptor has been demonstrated, with punctate alpha bungarotoxin staining on the ioSkeletal Myocyte surface in the presence of ioMotor Neurons.
